Calponin is a calmodulin, F-actin and tropomyosin binding protein, which is thought to be involved in the regulation of smooth muscle contraction. Calponin expression is restricted to smooth muscle cells and has been shown to be a marker of the differentiated (contractile) phenotype of developing smooth muscle. Multiple isoelectric variants of calponin have been identified, however only two molecular weight isoforms exist, a 34 kDa form and a 29 kDa form. Expression of the 29 kDa form, I-calponin, is primarily restricted to muscle of the urogenital tract, whereas the higher molecular weight variant has been demonstrated in vascular and visceral smooth muscle. In Western blotting, this MAb reacts with only the 34 kDa form of calponin in extracts of human aortic medial smooth muscle and is unreactive with fibroblast extracts of cultivated human foreskin. Beta-catenin associates with the cytoplasmic portion of E-cadherin. The catenin/cadherin complexes play an important role mediating cellular adhesion, including adherens junctions. Beta-catenin is involved in the Wnt signaling pathway as well as other signaling pathways, and is also found in complexes with many different proteins including the tumor suppressor protein APC. Defects in beta-catenin are associated with colorectal cancer, as well as many other cancer types. Latent or complexed PAI-1 will not bind to the plate and will not be detected. After appropriate washing steps, polyclonal anti-mouse PAI-1 primary antibody binds to the captured enzyme. Excess antibody is washed away and bound polyclonal antibody is then reacted with the secondary antibody conjugated to horseradish peroxidase. TMB substrate is used for color development at 450nm. Color development is directly proportional to the concentration of active PAI-1 in the samples. A standard calibration curve is prepared along with the samples to be measured using dilutions of mouse PAI-1.

This mucin is present in primary ovarian mucinous cancer but usually absent in colorectal adenocarcinoma, thus showing an expression pattern opposite to MUC2. This MAb is highly specific to p21 and shows no cross-reaction with other closely related mitotic inhibitors. p21WAF1 is a specific inhibitor of cdk s and a tumor suppressor involved in the pathogenesis of a variety of malignancies. The expression of this gene acts as an inhibitor of the cell cycle during G1 phase and is tightly controlled by the tumor suppressor protein p53.
